Synaptic Input Stimulation

Synaptic input stimulation is an experimental approach that activates presynaptic pathways to reproduce or manipulate the signals neurons receive at their synapses. Stimulation can use electrical pulses or light-sensitive proteins to trigger action potentials in selected axons, causing neurotransmitter release and measurable postsynaptic currents or potentials. In neuroscience, researchers use this method to examine synaptic transmission, characterize neural circuit connectivity, and measure how input strength and timing influence neuronal responses. Repeated or patterned stimulation can also reveal synaptic plasticity, including long-term potentiation and depression, providing insight into learning, memory, and circuit dysfunction.

Channelrhodopsin2 Mediated Stimulation of Synaptic Potentials at Drosophila Neuromuscular Junctions

This procedure uses a blue light-activated algal channel and cell-specific genetic expression tools to evoke synaptic potentials with light pulses at the neuromuscular junction (NMJ) in Drosophila larvae. This technique is an inexpensive and easy-to-use alternative to suction electrode stimulation for synaptic physiology studies in research and teaching laboratories.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

This video shows the craniotomy procedure that allows chronic imaging of neurons in mouse retrosplenial cortex using in vivo two photon microscopy in Thy1-GFP transgenic line. This approach is combined with injection of mCherry-expressing adeno-associated virus into dorsal hippocampus. These techniques allow long-term monitoring of experience-dependent structural plasticity in RSC.
