Kidney Function

Kidney function encompasses the processes by which the kidneys filter blood, regulate body fluid composition, and maintain physiological balance, making it central to health and pharmacology. In the nephron, glomerular filtration removes water and small solutes from plasma, while tubular reabsorption and secretion selectively return substances to the blood or eliminate them in urine; hormonal signals help regulate electrolytes, acid-base status, and fluid volume. These processes determine how many drugs and metabolites are cleared from the body, influencing dosing, duration of action, and drug interactions. Assessing kidney function also helps identify nephrotoxicity and guide safer treatment for patients with renal impairment.

The zebrafish adult kidney is an excellent system for renal regeneration and disease studies. An essential aspect of such research is the assessment of nephron structure and function. This protocol describes several methodologies that can be implemented to assess nephron tubule composition and to evaluate renal reabsorption.

The zebrafish is a popular tool to model chronic kidney disease (CKD). However, their small size makes it impossible to evaluate renal function using traditional methods. We describe a fluorescent dye kidney clearance assay1 that allows quantitative analysis of zebrafish kidney function in CKD.
