Preclinical Efficacy

Preclinical efficacy is the evaluation of whether a drug candidate produces its intended biological effect before testing in humans, making it a central stage of pharmacological development. Researchers assess target engagement, dose-response relationships, biomarkers, and functional outcomes in cell-based assays and relevant animal models, often comparing treated groups with controls. These studies help determine pharmacological activity, identify promising compounds, guide dose selection, and reveal limitations in a candidate’s mechanism or model performance. By connecting molecular action to measurable effects in living systems, preclinical efficacy research supports evidence-based decisions about advancing therapies into clinical trials, while recognizing that preclinical success does not guarantee human benefit.

We describe methods for longitudinal monitoring of the efficacy of therapeutics for the treatment of colonic pathologies in mice using a rigid endoscope. This protocol can be readily used for the characterization of the therapeutic response of an individual tumor in live mice and also for monitoring potential disease relapse.
