Zellen können die Genexpression bei jedem Schritt, von der DNA bis zum Protein, präzise regulieren. Diese Regulation erfolgt während der Transkription; während der RNA-Verarbeitung, -Lokalisierung und -Degradation; sowie während und nach der Übersetzung. 
Die Transkriptionsregulation wird durch Proteine vermittelt, die an regulatorische Sequenzen auf der DNA binden. Diese Transkriptionsfaktoren sind eine der gebräuchlichsten Methoden zur Kontrolle der Genexpression und können die Transkription entweder initiieren oder verhindern.
Die Transkription erzeugt Prä-mRNA, die durch mehrere regulierte Prozesse zu reifer mRNA verarbeitet werden muss. 
mRNA-Spleißen, das die nicht-kodierenden Regionen in der Vorläufer-mRNA entfernt und die kodierenden verbindet, steuert die Genexpression durch differentielle Spleißmuster und RNA-bindende Proteine. Das Hinzufügen eines Poly-A-Schwanzes und einer 5'-Kappe zur Produktion der reifen mRNA wird ebenfalls kontrolliert.
Als nächstes muss sich die mRNA mit RNA-bindenden Proteinen verbinden, um einen Komplex zu bilden, der als Ribonukleopartikel bekannt ist. Dieser Prozess ist hochgradig reguliert, und nur eine mRNA, die in einem Ribonukleopartikel vorliegt, kann vom Zellkern in das Zytoplasma transportiert werden, um translatiert zu werden.
Die Translationskontrolle ist ein weiterer entscheidender Punkt für die Regulation der Genexpression. Die Regulation kann spezifisch sein, wenn ein Individuum oder eine Untergruppe von mRNAs beteiligt ist, oder allgemein, wenn die meisten mRNA-Transkripte betroffen sind. 
Bei der spezifischen Regulation wird die Hemmung der Translation durch Wechselwirkungen mit transaktiven Elementen wie Proteinen und bestimmten RNA-Typen, einschließlich microRNAs und kurzen interferierenden RNAs, gesteuert.

Regulation of Expression Occurs at Multiple Steps

Die Genexpression kann nahezu auf jedem Schritt vom Gen zum Protein reguliert werden. Die Transkription ist der Schritt, der am häufigsten reguliert wird. Dies umfasst die Bindung von Proteinen an kurze regulatorische Sequenzen auf der DNA. Diese Assoziation kann entweder die Transkription eines mit der jeweiligen Sequenz assoziierten Gens fördern oder hemmen.

Die Transkription führt zur Bildung eines Vorläufers (Prä-mRNA), der sowohl Exons als auch Introns enthält und der weiteren Verarbeitung bedarf, bevor er in ein Protein übersetzt wird. Dies geschieht durch mRNA-Spleißen, das die Entfernung nicht kodierender Regionen und die Zusammenführung der kodierenden Bereiche umfasst. Die mRNA-Verarbeitung kann auch als regulatorischer Mechanismus durch Variationen in den Spleißmustern genutzt werden, wie das Überspringen bestimmter Exons, alternatives Spleißen und die Einbeziehung von Introns.

Das Hinzufügen eines Poly-A-Schwanzes am 3'-Ende und einer 5'-Kappe zur Erzeugung der reifen mRNA sind ebenfalls regulatorische Punkte während der RNA-Verarbeitung. Die Regulation erfolgt durch Variationen im Polyadenylierungssignal, das bestimmt, wo der Poly-A-Schwanz an die mRNA angefügt wird. In einigen Fällen ist mehr als ein Poly-A-Signal am 3'-Ende vorhanden, was die Länge der 3'-unübersetzten Region ändert, jedoch bleibt das Endprodukt des Proteins dasselbe. Die Stabilität und das Übersetzungspotenzial der mRNA-Varianten können jedoch unterschiedlich sein, was die Menge des produzierten Proteins verändern kann. In anderen Fällen ist ein zusätzliches Poly-A-Signal auf dem Intron oder Exon innerhalb der Gensequenz vorhanden, was zu Variationen in den Spleißstellen für die Polyadenylierung führen und unterschiedliche Proteine aus demselben Strang der Prä-mRNA erzeugen kann. Die Hinzufügung der 5'-Kappe, die aus methyliertem Guanosin besteht, wird durch zwei Mechanismen reguliert. Einer betrifft die Regulation der Methyltransferasen, die die Methylgruppe an das Guanosin anfügen, der andere die Regulation der zellulären Signalwege, die zur Methylierung führen.

Als Nächstes muss die reife mRNA durch Kernporenkomplexe (NPCs) vom Zellkern in das Zytoplasma transportiert werden, um übersetzt zu werden. Dies wird durch die mRNA reguliert, die einen Komplex bildet, bekannt als das Ribonukleoprotein, mit RNA-bindenden Proteinen. NPCs lassen nur mRNAs, die sich im Komplex befinden, in das Zytoplasma passieren. Sobald eine mRNA in das Zytoplasma zur Übersetzung gelangt, kann sie entweder individuell oder als Teil einer Gruppe durch spezifische Regulationen gezielt werden, oder sie kann einer gemeinsamen Regulation mit allen anderen mRNAs im Zytoplasma unterzogen werden. Bei spezifischer Regulation regulieren bestimmte trans-aktive Elemente, wie Proteine und verschiedene Arten von RNAs, die Transkription. Bei allgemeiner Regulation werden die Proteine, die an der Translationsmaschinerie beteiligt sind, aktiviert oder gehemmt, was wiederum die Übersetzung aller Transkripte beeinflusst. Der häufigste regulatorische Mechanism bei der Translation ist die Modifikation des Initiationsfaktors der Translation.

Die Genexpression kann auch durch posttranslationale Modifikationen reguliert werden, bei denen eine enzymatisch katalysierte reversible Modifikation die Funktion eines Proteins verändern kann. Eine häufige posttranslationale Modifikation ist die Phosphorylierung, die von Enzymen, den sogenannten Kinasen, durchgeführt wird. Die Dephosphorylierung von Proteinen wird andererseits von Proteinen, den sogenannten Phosphatasen, durchgeführt. Die Phosphorylierung eines Proteins kann zu dessen Aktivierung oder Deaktivierung führen und dessen Funktion ändern.

Cells control gene expression at several points between DNA and protein. The most common control point is transcription. At this stage, proteins bind to short regulatory DNA sequences and can either increase or reduce transcription of the linked gene.

After transcription, a precursor mRNA is made. This pre-mRNA contains exons, which code for protein, and introns, which do not. It must be processed before translation, and splicing removes the introns and joins the exons. Cells can also regulate gene expression through changes in splicing patterns, including skipping certain exons, alternative splicing, and including introns.

RNA processing also includes adding a 3' poly-A tail and a 5' cap to form mature mRNA. The poly-A tail is added at a site marked by a polyadenylation signal. In some cases, more than one poly-A signal is present at the 3' end. This can change the length of the 3' untranslated region without changing the final protein, but the mRNA variants may differ in stability and translation potential. In other cases, an extra poly-A signal in an intron or exon can change splicing during polyadenylation and produce different proteins from the same pre-mRNA strand.

The 5' cap is a methylated guanosine, and its addition is also regulated. One control route acts through methyltransferases, the enzymes that add the methyl group. Another route works through cellular signaling pathways that lead to methylation. After processing, mature mRNA must leave the nucleus through nuclear pore complexes. It does this by forming a ribonucleoparticle with RNA-binding proteins, and only mRNAs in this complex can pass into the cytoplasm.

Once mRNA reaches the cytoplasm, translation can be regulated in specific ways or in a more general way. Specific regulation affects particular transcripts through trans-acting elements such as proteins and different types of RNA. General regulation affects all cytoplasmic mRNAs by turning translation machinery proteins on or off. The most common translational control step is modification of the translation initiation factor. Gene expression can also change after a protein is made through post-translational modifications. A common example is phosphorylation, which is carried out by kinases, while phosphatases remove the phosphate group through dephosphorylation. Phosphorylation can activate or deactivate a protein and change its function.
