DNA Isolation

Für viele Anwendungen, wie z. B. die rekombinante DNA-Technologie, muss DNA aus Zellen isoliert und an präzisen Positionen geschnitten werden.
Obwohl für verschiedene Zelltypen unterschiedliche Arten von DNA-Extraktionsmethoden verwendet werden, gibt es drei Standardschritte: Zelllyse, Proteinentfernung und DNA-Rückgewinnung.
In eukaryotischen Zellen befindet sich die DNA im Zellkern. Daher müssen sowohl die Zellmembran als auch die Kernmembran aufgebrochen werden, um die DNA zu isolieren.
Dieser Schritt kann mechanisch erfolgen, indem Zellen durch Mahlen oder Beschallung aufgebrochen werden, oder chemisch durch die Verwendung von Detergenzien und Enzymen, um Teile der Zellmembran aufzulösen.
Sobald der Zellinhalt freigesetzt wird, werden die Ablagerungen durch Zentrifugation von den löslichen Bestandteilen getrennt. Der zurückgewonnene Überstand enthält Nukleinsäuren und wasserlösliche Proteine.
Um Proteine zu entfernen, werden dem Überstand Enzyme wie Proteinase K, Peptidase oder Lysozym zugesetzt, um die Peptidbindungen aufzubrechen.
Die DNA wird durch Fällung durch Zugabe von Alkohol und einem Salz, wie z. B. Natriumacetat, aus dem Überstand gewonnen.

DNA-Isolierungsprotokolle können schnell und einfach oder komplex und zeitaufwändig sein, abhängig von der Art und Qualität der DNA, die für die weitere Verarbeitung benötigt wird. Zum Beispiel ist die Extraktion von Plasmid-DNA etwas komplizierter als die von genomischer DNA, da eine geeignete Lysemethode erforderlich ist, um Plasmid-DNA von gDNA während der Isolation zu trennen. Für spezielle Anwendungen, wie die Sequenzierung von DNA über große Distanzen, die eine gute Ausbeute an hochwertigen DNA-Proben erfordern, müssen wir auch für die Extraktion von genomischer DNA spezifische Protokolle befolgen.

Arten von Methoden zur Extraktion von genomischer DNA

Das Hauptziel der Methoden zur Extraktion von genomischer DNA ist es, gDNA von Proteinen, RNA und anderen Zellinhalten zu trennen. Dies umfasst vier grundlegende Schritte: 1. Mechanische oder chemische Zerstörung der Zellstruktur zur Gewinnung des Zelllysats. 2. Schutz der DNA vor Abbau während der Verarbeitung. 3. Trennung der löslichen DNA von Zelltrümmern. 4. Elution der gereinigten DNA.

Die meisten Protokolle zur Isolierung von genomischer DNA basieren entweder auf lösungsbasierten oder festphasenbasierten Extraktionsmethoden. Lösungsbasierte Methoden setzen auf Fällung und Zentrifugation, um DNA von anderen zellulären Materialien zu trennen, gefolgt von organischer Extraktion oder "Aussalzung", um lösliche DNA von zellulären Proteinen zu trennen. Die abschließende DNA-Fällung erfolgt mit Ethanol. Genomic DNA extraction methods separate DNA from proteins, RNA, and other cell material. The process usually follows four main steps. First, the cell structure is disrupted with mechanical or chemical methods to make a cell lysate. Then the DNA is protected from damage during processing. After that, the soluble DNA is separated from cell debris. Finally, the purified DNA is eluted, or washed out, for later use.

DNA isolation can follow two main approaches: solution-based methods or solid-phase extraction methods. Solution-based methods use precipitation and centrifugation to separate DNA from other cellular material. They may also use organic extraction or salting out to remove proteins from the soluble DNA. The final DNA is then precipitated with ethanol.

Solid-phase extraction methods use a solid support, such as silica or cellulose matrices, to bind the DNA. The sample is then washed so other cellular components are removed. Next, the DNA is eluted from the solid support. These methods may use centrifugation, vacuum, or magnetic steps to separate the bound DNA during the process.

The best DNA extraction method depends on the sample type, how many samples must be processed, and the next step for the DNA. Plasmid DNA extraction is usually more complex than genomic DNA extraction because the lysis method must separate plasmid DNA from genomic DNA during isolation. In some cases, such as long-range DNA sequencing, specific protocols are needed to obtain enough high-quality DNA.
