Bei der Immungold-Elektronenmikroskopie wird ein Zielmolekül mit goldkonjugierten Antikörpern markiert.
Das Gold erhöht die Elektronenstreuung, so dass kontrastreiche dunkle Flecken entstehen. Diese Spots können von den anderen unmarkierten Strukturen unterschieden werden und ermöglichen die Lokalisierung des Zielmoleküls.
Bei der direkten Immunmarkierung wird eine Goldsonde verwendet, die an einen primären Antikörper gebunden ist, der direkt an das Zielmolekül bindet. 
Bei der indirekten Markierung wird ein goldmarkierter Sekundärantikörper verwendet, der an einen unmarkierten Primärantikörper bindet. Die indirekte Markierung wird aufgrund ihrer höheren Empfindlichkeit häufiger verwendet.
Die Immunmarkierung kann mit verschiedenen Techniken durchgeführt werden – Post-Embedding, Pre-Embedding oder Whole-Mount.
Bei der Post-Embedding-Technik wird eine Probe in Harz eingebettet und dann in ultradünne Schnitte geschnitten, die dann mit goldmarkierten Antikörpern behandelt werden.
Bei der Pre-Embedding-Technik wird eine Probe zunächst mit den goldmarkierten Antikörpern inkubiert und anschließend in ein Harz eingebettet und mit einem Querschnitt versehen.
Bei der Whole-Mount-Technik wird kein Harz eingebettet. Antikörperreaktionen werden direkt an einer Probe durchgeführt, die auf dem Probenhalter platziert wird.

Immunogold Electron Microscopy

Die Immunelektronenmikroskopie nutzt die Immunogold-Markierung endogener Proteine mit spezifischen Antikörpern, um diese Proteine in Zellen und Geweben nachzuweisen und zu lokalisieren. Das Verfahren liefert Einblicke in die Verteilung und Quantifizierung von Proteinen unter verschiedenen Stimulationsbedingungen und gibt Hinweise auf ihre Funktionen. Die Konjugation hochelektronendichter Goldpartikel mit primären oder sekundären Antikörpern ermöglicht den Antigennachweis auf und innerhalb von Zellen mit hoher Auflösung und Spezifität. Immunelektronenmikroskopie wurde verwendet, um die zelluläre und subzelluläre Lokalisierung von Proteinen zu identifizieren, die an der Neurotransmission beteiligt sind, Kernproteinkomponenten und die Identifizierung von Immunzelltypen.

Zur Lokalisierung von Zellantigenen mittels Transmissionselektronenmikroskopie werden drei Ansätze angewendet. Erstens umfassen die Protokolle bei der Untersuchung der Lokalisierung intrazellulärer Antigene die Antikörpermarkierung nach dem Einbetten in Acrylharze, die Antikörpermarkierung vor dem Einbetten in das Harz in Kombination mit der Permeabilisierung der Zellmembran oder die Kryo-Ultramikrotomie ohne Einbettung. Zweitens wird bei der Lokalisierung von Zelloberflächenproteinen das Voreinbettungsprotokoll verwendet, bei dem die Markierung mit Immunogold-Antikörpern vor der Einbettung in das Harz erfolgt. Bei der dritten Technik, der Whole-Mount-Technik, ist keine Harzeinbettung erforderlich, und die Antikörperreaktionen werden direkt an einer Probe durchgeführt, um die Oberflächenmoleküle zu lokalisieren.

Für die Rasterelektronenmikroskopie muss die interessierende Oberfläche unbedingt freigelegt werden. Wenn die äußere Oberfläche der Plasmamembran von Interesse ist, kann die Probe direkt fixiert und markiert werden. Allerdings müssen die Zellen mit Detergentmoleküleien permeabilisiert und immunmarkiert werden, um intrazelluläre Komponenten oder in ein Gewebe integrierte Antigene zu untersuchen. Zur Betrachtung wird die Probe gefriergebrochen (schnelles Einfrieren und Brechen mit einem Messer). Viele intrazelluläre Komponenten, insbesondere makromolekulare Komplexe wie Chromatin, Proteinkomplexe oder Viren, können für die SEM isoliert, fixiert und immunmarkiert werden. Abschließend wird eine immunnegative Färbetechnik für Oberflächenantigene auf kleinen Proben wie Viren und Bakterien angewendet, die sich für eine negative Färbung eignen.

Immunogold labeling in electron microscopy uses antibodies and tiny gold particles to find specific proteins in cells and tissues. The gold particles are very electron-dense, which means they show up clearly in electron microscope images. This gives researchers a high-resolution way to see where proteins are located and to compare their amounts under different stimulation conditions.

The method can reveal proteins on the cell surface and inside cells. It has been used to study proteins involved in neurotransmission, parts of the nucleus, and markers for immune cell types. The same labeling idea can be paired with different sample-preparation methods, depending on what needs to be seen.

For transmission electron microscopy, three main approaches are used to localize cell antigens. Intracellular antigens can be labeled after embedding in acrylic resin, before embedding with membrane permeabilization, or by cryo-ultramicrotomy without embedding. When cell-surface proteins are the target, pre-embedding labeling is used so the immunogold antibodies bind before the sample is placed in resin.

A whole-mount technique is also used. In this method, the sample is not embedded in resin, and the antibody reaction happens directly on the specimen to identify surface molecules. For scanning electron microscopy, the surface of interest must be exposed. The sample can be fixed and labeled directly if the outer surface of the plasma membrane is being studied.

If intracellular components or antigens within a tissue are being examined with SEM, the cells must be permeabilized with detergents and then immunolabeled. The sample is then freeze-fractured, which means it is rapidly frozen and broken with a knife for viewing. Many intracellular components, including chromatin, protein complexes, and viruses, can be isolated, fixed, and immunolabeled this way.

An immunonegative staining technique is used for small specimens such as viruses and bacteria. This method is suited to surface antigens on samples that work well with negative staining. Together, these approaches let scientists localize proteins and other antigens with strong specificity and fine structural detail.
