Die 3D-Struktur biologischer Proben kann mit zwei Kryo-Elektronenmikroskopie-Techniken bestimmt werden – der Elektronentomographie und der Einzelpartikelrekonstruktion.
Bei der Elektronentomographie wird eine Probe relativ zum Elektronenstrahl progressiv gekippt, wodurch eine Reihe von 2D-Bildern aus verschiedenen Winkeln aufgenommen wird. Diese 2D-Bilder werden dann mit Hilfe einer Computersoftware gestapelt, um das 3D-Bild oder Tomogramm der Probe zu erzeugen.
Die Elektronentomographie erhält die strukturelle Integrität der Probe in vivo und wird hauptsächlich für die 3D-Visualisierung von Zellen, Organellen und Makromolekülen in ihrer natürlichen Umgebung verwendet.
Bei der Einzelpartikelrekonstruktion werden isolierte in vitro Proben, wie z. B. eine gereinigte Proteinprobe, gleichmäßig auf einem Gitter verteilt.
Es wird ein 2D-Bild der einzelnen Proteinmoleküle aufgenommen, die in zufälligen Orientierungen liegen. Tausende solcher Bilder werden dann gemittelt und ausgerichtet, um das 3D-Bild der Probe zu rekonstruieren.

Electron Microscope Tomography and Single-particle Reconstruction

Mithilfe der Transmissionselektronenmikroskopie (TEM) kann mithilfe von Techniken wie der Elektronenmikroskop-Tomographie und der Einzelpartikelrekonstruktion die 3D-Struktur biologischer Proben bestimmt werden. Während die Einzelpartikelrekonstruktion Makromoleküle und makromolekulare Komplexe nur unter In-vitro-Bedingungen untersuchen kann, ermöglicht die Tomographie die Untersuchung von Zellbestandteilen oder kleinen Zellen in vivo.

Elektronentomographie

Die Elektronentomographie kann entweder im TEM- oder STEM-Modus (Rastertransmissionselektronenmikroskopie) durchgeführt werden. STEM ist eine Technik, die hauptsächlich zur Erstellung von Tomogrammen dicker biologischer Proben verwendet wird. Es kombiniert die Probenoberflächen-Scanmethode der Rasterelektronenmikroskopie, sodass die an jedem Punkt, an dem der Elektronenstrahl auf die Probe trifft, durchgelassenen oder gestreuten Elektronen von einer Reihe von Detektoren gesammelt werden.

Eine Variante der herkömmlichen Elektronentomographietechnik ist die Zweiachsentomographie, bei der die Probe gegenüber dem Elektronenstrahl um zwei Achsen geneigt wird. Dies führt zu zwei separaten Tomogrammen, die dann ausgerichtet werden, um ein 3D-Bild der Probe zu erzeugen. Die zweiachsige Tomographie hat gegenüber der einachsigen Tomographie zwei Vorteile: eine bessere Rekonstruktion der erweiterten Merkmale der Probe und eine höhere Tiefenauflösung der Probe.

Eine weitere Variante ist die Elektronen-Kryo-Tomographie, bei der Proben unter kryogenen Bedingungen abgebildet werden, da durch Fixierung und Austrocknung die biologischen Strukturen beschädigt werden können. Die Methode wird vor allem bei dünnen Proben mit einer Dicke von weniger als 500 nm eingesetzt, da dicke Proben den Elektronenstrahl blockieren. Daher war die Technik auf gereinigte makromolekulare Komplexe, Viren oder kleine Zellen wie Bakterienzellen beschränkt.

Einzelpartikelrekonstruktion

Die Einzelpartikelanalyse wird typischerweise in der Kryo-Elektronenmikroskopie verwendet, um 3D-Strukturen mit nahezu atomarer Auflösung zu erzeugen. Es eignet sich am besten für große oder dynamische makromolekulare Komplexe, die schwer zu kristallisieren sind, und ist daher ein Ersatz für die Röntgenkristallographie. Für kristallisierbare Komplexe ist die Einzelpartikelanalyse eine Methode der Wahl, um hochauflösende Strukturen zu erhalten. Die Technik wurde zur Untersuchung von Membranproteinen, Proteinkomplexen, der Chromatinstruktur und makromolekularen Maschinen wie Ribosomen und Proteasomen eingesetzt.

Transmission electron microscopy (TEM) can build 3D images of biological samples. It does this with electron microscope tomography and single-particle reconstruction. Tomography can show cell parts or small cells in vivo, while single-particle reconstruction is used for macromolecules and macromolecular complexes in vitro only.

Electron tomography can be done in TEM or in STEM, which stands for scanning transmission electron microscopy. STEM is mainly used for thick biological specimens. It combines sample scanning with detectors that collect electrons transmitted or scattered where the beam hits the sample.

A common variation is dual-axis tomography. In this method, the sample is tilted around two axes relative to the electron beam. The two tomograms are then aligned to form a 3D image. Dual-axis tomography can improve the reconstruction of expanded features and increase depth resolution compared with single-axis tomography.

Another form is electron cryo-tomography. Samples are imaged under cryogenic conditions because fixation and dehydration can damage biological structures. This method is mainly used for thin samples, usually less than 500 nm thick, because thicker samples block the electron beam. It has therefore been limited to purified macromolecular complexes, viruses, and small cells such as bacterial cells.

Single-particle analysis is usually paired with cryo-electron microscopy to produce 3D structures with near-atomic resolution. It works best for large or dynamic macromolecular complexes that are hard to crystallize, so it can serve as an alternative to X-ray crystallography. When complexes can be crystallized, single-particle analysis is still a method of choice for obtaining high-resolution structures. It has been used to study membrane proteins, protein complexes, chromatin structure, and macromolecular machines such as ribosomes and proteasomes.
