Human Peripheral Blood Neutrophils

Human peripheral blood neutrophils are mature, short-lived white blood cells that provide rapid innate immune defense and serve as a useful model for studying inflammatory biochemistry. In response to chemotactic signals, they migrate toward sites of tissue damage or infection, recognize and engulf particles, and eliminate them through granule-mediated enzymes, reactive oxygen species, and neutrophil extracellular traps. Researchers isolate these cells from blood to measure phagocytosis, degranulation, oxidative bursts, and signaling pathways, supporting investigations of host-pathogen interactions, acute inflammation, autoimmune disease, and potential therapeutic targets. Their biochemical responses also help clarify how immune activation contributes to tissue injury and disease.

This protocol allows for the reliable generation and characterization of blood outgrowth endothelial cells (BOECs) from a small volume of adult peripheral blood. BOECs can be used as a surrogate for endothelial cells from patients with vascular disorders and as a substrate for the generation of induced pluripotent stem cells.
