Plasma Membrane Diffusion

Plasma membrane diffusion is the passive movement of molecules across a cell’s selectively permeable membrane, a process that helps maintain chemical balance and support cellular function. Molecules move down their concentration or electrochemical gradients without direct ATP use; small nonpolar substances can pass through the lipid bilayer, while ions and polar molecules typically require channel or carrier proteins for facilitated diffusion. In biochemistry, analyzing diffusion clarifies membrane permeability, transport specificity, osmotic behavior, and cellular responses to changing environments. These principles also inform studies of drug delivery, membrane protein activity, and the molecular basis of homeostasis.

Polarization-based Total Internal Reflection Fluorescence Microscopy (pTIRFM) enables real-time detection of cell membrane dynamics. This article describes the implementation of pTIRFM for the study of membrane remodeling during regulated exocytosis. The technique is generalizable to other processes in cell biology that directly or indirectly involve changes in membrane shape.
