Background Subtraction

Background subtraction is an image- and video-processing method that separates objects of interest from relatively static surroundings, making it useful for analyzing biological scenes. It typically builds a background model from reference images or temporal pixel statistics, then subtracts the estimated background from each new frame; thresholding the resulting differences produces a foreground mask. In bioengineering, this approach can isolate cells, organisms, particles, or tissue features in microscopy and time-lapse recordings, supporting segmentation, motion tracking, and quantitative measurements. Careful handling of illumination changes, noise, and gradual background motion improves detection reliability and helps convert complex images into interpretable biological data.

We discuss the construction and operation of a complex nonlinear optical system that uses ultrafast all-optical switching to isolate Raman from fluorescence signals. Using this system we are able to successfully separate Raman and fluorescence signals utilizing pulse energies and average powers that remain biologically safe.
