Blastocyst Re-expansion

Blastocyst re-expansion is the recovery and renewed enlargement of the fluid-filled cavity within a blastocyst after cryopreservation and warming, indicating that the embryo has resumed normal cellular activity. During vitrification, rapid cooling preserves the embryo in a glass-like state; after warming, surviving trophectoderm cells restore membrane transport and fluid accumulation, causing the blastocoel to expand again. In bioengineering and assisted reproductive research, monitoring re-expansion helps assess embryo survival and post-thaw quality before further culture or transfer. The process also supports the development of improved cryoprotective solutions, warming protocols, imaging methods, and embryo assessment systems.

Blastocyst biopsy and vitrification are required to efficiently conduct preimplantation genetic testing. An approach entailing the sequential opening of the zona pellucida and retrieval of 7-8 trophectoderm cells in day 5-7 post-insemination limits both the number of manipulations required and the exposure of the embryo to sub-optimal environmental conditions.
