Branched-dna Technology

Branched-DNA technology is a nucleic acid detection method that measures specific RNA or DNA sequences by amplifying the signal generated from target hybridization rather than copying the target itself. In a typical assay, capture probes immobilize the target, extender probes recruit branched-DNA amplifier molecules, and multiple labeled probes bind to each amplifier to produce a measurable chemiluminescent signal. This architecture improves detection of low-abundance transcripts while preserving sequence specificity and simplifying quantitative analysis. In bioengineering, branched-DNA assays support gene-expression profiling, pathogen or viral-load measurement, and evaluation of engineered cells, providing a scalable alternative to target-amplification methods.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.

This protocol describes high-throughput plasmid transfection of mammalian cells in a 384-well plate using acoustic droplet ejection technology. The time-consuming, error-prone DNA dispensing and multiplexing, but also the transfection reagent dispensing, are software-driven and performed by a nanodispenser device. The cells are then seeded in these prefilled wells.
