Ficoll 70

Ficoll 70 is a highly branched, water-soluble polysucrose polymer with an average molecular mass of about 70 kDa, used to control solution density, viscosity, and macromolecular crowding in biological systems. In density-gradient centrifugation, its polymer concentration produces a defined medium in which cells or particles migrate according to buoyant density, while its excluded-volume effects can increase effective concentrations of biomolecules in solution. Bioengineers use Ficoll 70 in cell-separation workflows, biomimetic crowded environments, and in vitro studies of biochemical reactions and assembly. These applications support cell processing, tissue engineering research, and the design of physiologically relevant experimental systems.

Here, a protocol involving re-arterialized rat partial liver transplantation is presented. Specifically, 70% liver was resected in vivo by using an updated technique of vessel-oriented hepatectomy. The hepatic artery was reconstructed in an end-to-side manner. The cuff technique was modified to shorten the anastomosis time of the infrahepatic vena cava.

This protocol demonstrates the chaperone activity of heat shock protein 70 (Hsp70). E. coli dnaK756 cells serve as a model for the assay as they harbor a native, functionally impaired Hsp70, making them susceptible to heat stress. The heterologous introduction of functional Hsp70 rescues the growth deficiency of the cells.
