Placental Chondroitin Sulfate Binding

Placental chondroitin sulfate binding is the molecular recognition of chondroitin sulfate glycans in placental tissue by proteins, cells, or engineered ligands, a process relevant to placental biology and disease research. Binding depends on complementary molecular features, including glycan sulfation patterns, charge, spacing, and the three-dimensional presentation of binding sites, which can be measured with biochemical assays or recreated on functionalized surfaces. In bioengineering, these interactions support targeted delivery systems, affinity-based sensors, and tissue-mimetic materials while helping researchers study placental cell adhesion and pathogen sequestration. Characterizing binding strength and specificity can improve diagnostic and therapeutic strategies.

Here, we present a protocol for the synthesis of placental chondroitin sulfate A binding peptide (plCSA-BP)-conjugated lipid-polymer nanoparticles via single-step sonication and bioconjugate techniques. These particles constitute a novel tool for the targeted delivery of therapeutics to most human tumors and placental trophoblasts to treat cancers and placental disorders.

An injectable tissue engineering scaffold composed of poly(N-isopropylacrylamide)-graft-chondroitin sulfate (PNIPAAm-g-CS)-containing alginate microparticles was prepared. The adhesive strength, swelling properties and in vitro biocompatibility are analyzed in this study. The characterization techniques developed here may be applicable to other thermogelling systems.

In this video, we demonstrate a procedure for combined Alcian blue and silver staining of sulfated glycosaminoglycan oligosaccharide fragments. This combined staining technique significantly amplifies the bands of glycosaminoglycan polymers of different lengths in polyacrylamide gels.

We present a detailed method to study human placental physiology in vivo at term. The method combines blood sampling from the incoming and outgoing vessels on the maternal and fetal sides of the placenta with ultrasound measurements of volume blood flow and placental tissue sampling.
