Protein-targeting Drug Screening

Protein-targeting drug screening is the systematic evaluation of chemical or biological compounds for their ability to bind, modify, or regulate a specific protein, a central step in developing therapeutics and bioengineering applications. Screening assays expose purified proteins or engineered cells to candidate molecules and measure changes in binding, enzymatic activity, signaling, or protein function to identify promising hits. Researchers then characterize these hits for potency, selectivity, and reproducibility, often refining their structures or delivery systems through bioengineering approaches. The resulting data support target validation, drug optimization, and the design of more precise treatments for disease.

A Rapid and Quantitative Fluorimetric Method for Protein-Targeting Small Molecule Drug Screening

A protocol for small molecular drug screening based on in-situ synthesis of ultrasmall fluorescent gold nanoclusters (Au NCs) using drug-loaded protein as template is presented. This method is simple to determine the binding affinity of drugs to a target protein by a visible fluorescent signal emitted from the protein-templated Au NCs.

Fission Yeast as a Platform for Antibacterial Drug Screens Targeting Bacterial Cytoskeleton Proteins

Fission yeast is used here as a heterologous host to express bacterial cytoskeletal proteins such as FtsZ and MreB as translational fusion proteins with GFP to visualize their polymerization. Also, compounds that affect polymerization are identified by imaging using a fluorescence microscope.
