Endothelial Cell Growth Factors

Endothelial cell growth factors are signaling proteins that regulate the growth, survival, migration, and specialization of endothelial cells lining blood vessels. Factors such as vascular endothelial growth factor and fibroblast growth factor bind receptors on endothelial cells, activating intracellular pathways that promote cell proliferation, directed migration, and new vessel formation through angiogenesis. In biological techniques, these factors support endothelial cell culture, vascular tissue engineering, wound-healing studies, and assays of angiogenic activity. Controlling their concentration and timing helps researchers model vascular development, investigate disease mechanisms, and evaluate therapies that either stimulate or inhibit blood-vessel growth.

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
