Phenyl Agarose Resin

Phenyl agarose resin is a chromatography medium used to separate and purify proteins according to differences in surface hydrophobicity. It consists of porous agarose beads bearing phenyl groups, which interact with hydrophobic regions of proteins when high concentrations of salts such as ammonium sulfate reduce water availability and promote hydrophobic association. As the salt concentration decreases, bound proteins elute in order of interaction strength, allowing selective fractionation under generally non-denaturing conditions. In biological techniques, phenyl agarose resin supports protein purification, sample cleanup, and separation of native protein forms while helping researchers prepare samples for biochemical or structural analyses.

Source: Schaeffer, J. W., Chandler, et al. Detection of Viruses from Bioaerosols Using Anion Exchange Resin. J. Vis. Exp. (2018).This video demonstrates the detection of viruses from bioaerosols using an anion exchange resin. A liquid impinger containing the resin is used to capture viruses from the air. The resin is collected and treated with a virus lysis buffer containing carrier RNA. The lysate is transferred to a microcentrifuge tube for viral RNA isolation, followed by qRT-PCR to amplify...

Immunoprecipitation-Based Techniques: Purification of Endogenous Proteins Using Agarose Beads

2023

Source: Susannah C. Shissler1, Tonya J. Webb1 1 Department of Microbiology and Immunology, University of Maryland, Baltimore, MD 21201 Immunoprecipitation (IP, also known as a 'pull-down' assay) is a widely used technique that has applications in a variety of fields. First conceived in 1984, it was refined in 1988 (1, 2). The fundamental goal of IP is purification and isolation of a specific protein using an antibody against that protein. The word "immuno" refers to the use of an antibody while...
