Aspergillus Fumigatus Asthma Model

The Aspergillus fumigatus asthma model is an experimental system that reproduces key features of allergic airway disease caused by immune responses to the environmental fungus A. fumigatus. In common animal models, sensitization followed by inhalation or airway exposure to fungal spores or extracts activates type 2 inflammation, leading to immunoglobulin E production, eosinophil recruitment, mucus secretion, and airway hyperresponsiveness. Researchers use this model to examine interactions among fungal antigens, airway epithelium, and adaptive immunity, while evaluating potential treatments for allergic asthma. It also helps distinguish inflammatory pathways involved in fungus-associated respiratory disease and supports development of targeted therapies.

Here, we describe a protocol to assess antifungal activity of primary human immune cells in real-time using fluorescent Aspergillus reporter conidia in conjunction with live-cell video microscopy and flow cytometry. Generated data provide insight into host cell-Aspergillus interactions such as fungicidal activity, phagocytosis, cell migration and inhibition of fungal growth.
