Atmospheric Oxygen

Atmospheric oxygen is the oxygen gas present in a planet’s atmosphere, where it supports aerobic life and influences Earth’s chemical environment. On Earth, photosynthetic organisms release oxygen by splitting water during photosynthesis, while animals, plants, fungi, and many microorganisms consume it during cellular respiration, using oxygen as the terminal electron acceptor in the mitochondrial electron transport chain to generate ATP. Oxygen availability shapes metabolism, ecosystems, decomposition, and organism distribution, while its atmospheric concentration also contributes to ozone formation and protection from ultraviolet radiation. Studying atmospheric oxygen helps biologists understand evolution, energy flow, and the environmental conditions that sustain life.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
