Cotton Seedling Growth

Cotton seedling growth is the early developmental process through which a cotton seed germinates and forms a young plant, making it an important model for studying plant biology and crop establishment. After absorbing water, the seed activates metabolism, produces a radicle, and extends the hypocotyl; as cotyledons emerge and begin photosynthesis, the seedling shifts from dependence on stored reserves to self-supported growth. Measuring emergence, height, root development, and biomass helps researchers assess seed vigor and examine how temperature, moisture, light, soil conditions, and other environmental factors influence development. This knowledge supports biology research and agricultural practices aimed at improving stand establishment and early plant performance.

We present the detailed protocol for Agrobacterium-mediated virus-induced gene silencing (VIGS) assay in cotton. The tobacco rattle virus (TRV)-derived VIGS vectors were deployed to induce RNA silencing of cotton GrCLA1, Cloroplastos alterados 1 gene. The albino phenotype caused by silencing GrCLA1 was observed at the seedling stage within 2 weeks after inoculation.

Source: Hassan, J. A., et al. High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay. J. Vis. Exp. (2020).This video demonstrates a method to screen for bacterial resistance in different tomato strains using ten-day-old seedlings inoculated with a bacterial suspension. It explains how bacteria invade through stomata, deliver effector proteins to suppress plant immunity, and how plant responses differ between susceptible and resistant...

Fabricating Cotton Analytical Devices

2016

To investigate simple fabrication approaches for multiple assay needs, we created a fluid-absorbing channel system made of cotton material. This device was used to establish a multiple detection platform, and solve contamination issues that commonly affect lateral flow-based biomedical devices, for clinical urinalysis of nitrite, total protein, and urobilinogen.

This article demonstrates how to culture Arabidopsis thaliana seedlings in a two-layer microfluidic platform that confines the main root and root hairs to a single optical plane. This platform can be used for real-time optical imaging of fine root morphology as well as for high-resolution imaging by other means.
