Drosophila Brain Mounting

Drosophila brain mounting is a laboratory technique that prepares dissected fruit fly brains for microscopic examination while preserving their structure and orientation. After dissection, the tissue is typically fixed, stained or immunolabeled, and positioned in an aqueous or resin-based mounting medium beneath a coverslip; careful orientation and protection from compression help maintain fine neural anatomy. This preparation supports fluorescence, confocal, and other imaging methods used to examine neuronal morphology, brain regions, synaptic organization, and gene or protein expression. Reliable mounting improves image quality and enables comparisons of neural development, behavior-related circuitry, and disease models in biological research.

The adult Drosophila brain is a valuable system for studying neuronal circuitry, higher brain functions, and complex disorders. An efficient method to dissect whole brain tissue from the small fly head will facilitate brain-based studies. Here we describe a simple, one-step dissection protocol of adult brains with well-preserved morphology.
