Embryonic Day 12.5

Embryonic Day 12.5, often abbreviated E12.5, is a defined developmental timepoint approximately 12.5 days after fertilization in commonly used mouse models, when major embryonic structures undergo rapid growth and refinement. At this stage, coordinated cell proliferation, differentiation, migration, and tissue morphogenesis shape developing organs, while signaling interactions establish and maintain distinct tissue identities. Researchers use E12.5 embryos to examine organogenesis, congenital abnormalities, gene function, and responses to genetic or environmental perturbations. Because development progresses reproducibly by embryonic age, E12.5 provides a standardized reference for comparing anatomy, molecular expression, and developmental outcomes across experiments.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

This video shows a procedure for isolating intact protoplasts from tissues of 14-day-old seedlings of Arabidopsis. Given that the isolated protoplasts remain intact for at least 96h and are isolated from seedlings instead of one-month-old mature plants, this procedure expedites assays requiring intact protoplasts.
