Endothelium Dependent Vasodilation

Endothelium-dependent vasodilation is the widening of blood vessels caused by signals released from the endothelial cells lining the vascular wall, an essential process for regulating blood flow and vascular resistance. In response to stimuli such as increased shear stress or specific agonists, endothelial nitric oxide synthase produces nitric oxide, which diffuses into adjacent smooth muscle cells and activates guanylyl cyclase, increasing cyclic GMP and promoting relaxation. This mechanism supports cardiovascular homeostasis and provides a foundation for studying vascular physiology, endothelial function, and disorders associated with impaired blood-flow regulation, including atherosclerosis and hypertension.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.

Endothelial dysfunction is associated with numerous disease states and is predictive of adverse cardiovascular events in humans. Flow-mediated vasodilation (FMD) is a non-invasive ultrasound method of evaluating endothelial function. Methodological choices and operator experience may affect results. A systematic approach to FMD in human studies is discussed here.

Antihypertensive Drugs: Vasodilators

2024

Vasodilators, primarily affecting the smooth muscles within arterial and venous walls, are commonly used for hypertension treatment. Medications such as minoxidil and hydralazine primarily target arteries and arterioles, while sodium nitroprusside acts on arterioles and venules. Minoxidil, functioning as a prodrug, is metabolized by hepatic sulfotransferase into its active form, minoxidil sulfate, after oral administration. This metabolite binds to the sulfonylurea receptor (SUR) component of...
