High Density Lipoprotein

High-density lipoprotein (HDL) is a class of blood lipoprotein particles that transports cholesterol and other lipids through the body, supporting cellular lipid balance and vascular health. HDL particles acquire excess cholesterol from peripheral tissues, including macrophages, through membrane transporters such as ABCA1; the cholesterol is then esterified by lecithin–cholesterol acyltransferase and carried to the liver for processing or excretion. In biology, studying HDL structure, metabolism, and reverse cholesterol transport helps explain lipid homeostasis and cardiovascular disease mechanisms. HDL-related research also informs investigations of inflammation, atherosclerosis, and therapeutic strategies designed to improve lipoprotein function rather than simply increase HDL concentration.

Herein, we describe the protocols to measure the amount of fluorescent phospholipid specifically removed from lipid donor particles by high-density lipoprotein (HDL) present in whole human plasma or serum. We have shown that this metric of HDL functionality predicts incident cardiovascular disease.

MicroRNAs play an important regulatory role and are emerging as novel therapeutic targets for various human diseases. It has been shown that miRNAs are carried in high density lipoproteins. We have developed a simplified method to rapidly isolate purified HDL suitable for miRNA analysis from human plasma.
