Larval Dissection

Larval dissection is a biological technique for exposing and isolating internal tissues or organs from insect or other animal larvae so their structure and development can be examined. Under a stereomicroscope, researchers immobilize the larva, open the body wall with fine instruments, and remove surrounding tissues while preserving target structures for imaging, staining, or further analysis. This approach supports developmental biology by revealing how organs form, grow, and respond to genetic or environmental changes. It is also useful for studying anatomy, gene expression, neural and muscular organization, and disease-related phenotypes, providing direct cellular and tissue-level evidence that complements whole-organism observations.

How niches and stem cells form during development is an important question with practical implications. In the Drosophila ovary, germ line stem cells and their somatic niches form during larval development. This video demonstrates how to dissect, stain and mount female gonads from late third instar (LL3) Drosophila larvae.
