Myogenic Progenitor Cells

Myogenic progenitor cells are precursor cells that generate skeletal muscle, making them central to muscle development, repair, and regeneration. In response to developmental signals or tissue injury, these cells activate, proliferate, and commit to the myogenic lineage before differentiating into myoblasts that fuse into multinucleated myotubes and mature muscle fibers. Researchers study myogenic progenitor cells to understand muscle formation, satellite-cell activity, and regeneration, as well as disorders such as muscular dystrophy. Their capacity for self-renewal and muscle formation also supports applications in disease modeling, regenerative medicine, and tissue engineering.

Induced pluripotent stem cell (iPSC)-derived myogenic progenitors are promising candidates for cell therapy strategies to treat muscular dystrophies. This protocol describes transplantation and functional measurements required to evaluate the engraftment and differentiation of iPSC-derived mesoangioblasts (a type of muscle progenitors) in mouse models of acute and chronic muscle regeneration.

In vitro culture systems have proven indispensible to our understanding of vertebrate myogenesis. However, much remains to be learned about nonmammalian skeletal muscle development and growth, particularly in basal taxa. An efficient and robust protocol for isolating the adult stem cells of this tissue, the myogenic precursor cells (MPCs), and maintaining their self-renewal, proliferation, and differentiation in a primary culture setting allows for the identification of conserved and divergent...

Here, we demonstrate fabrication of collagen-based, tissue constructs containing skeletal myoblasts. These 3-D engineered constructs may be used to replace or repair tissues in vivo. For our purposes, we have designed these as an atrioventricular electrical conduit for the repair of complete heart block[1].

This protocol outlines a method to isolate Fibro-adipogenic progenitors (FAPs) and myogenic progenitors (MPs) from rat skeletal muscle. Utilization of the rat in muscle injury models provides increased tissue availability from atrophic muscle for the analysis and a larger repertoire of validated methods to assess muscle strength and gait in free-moving animals.
