Larval Instar Stages

Larval instar stages are the distinct growth phases that occur between successive molts in the immature forms of insects and other arthropods, providing a framework for describing development. During each instar, the larva feeds and increases in size, then sheds its rigid exoskeleton when hormonal signals, including ecdysone and juvenile hormone, trigger molting and determine whether development continues as a larva or progresses toward metamorphosis. Identifying instars helps biologists track growth, compare species, interpret life histories, and assess environmental effects on development. In applied biology, instar-specific knowledge supports insect identification, ecological research, agricultural pest management, and laboratory studies of development and metamorphosis.

Visualization of Larval Segmental Nerves in 3rd Instar Drosophila Larval Preparations

Drosophila melanogaster larvae provide an ideal model system to investigate the mechanisms of axonal transport within larval segmental nerves. Using this procedure, 3rd instar larvae carrying various mutations can be compared to wild type larvae.

This protocol demonstrates the dissection technique used for isolating imaginal discs from drosophila larvae at the 3rd instar stage. Methods for fixing the tissue after saying and removing the wing, leg, and eye discs are demonstrated directly on a microscope slide for subsequent visualization.
