Live Embryo Staining

Live embryo staining is a biological technique that uses dyes or fluorescent markers to visualize cells and structures in living embryos while development continues. The stain enters or labels specific cellular components, producing color or fluorescence that can be observed over time under suitable imaging conditions without immediately fixing the specimen. Researchers use this approach to follow cell division, migration, differentiation, tissue formation, and changes in embryonic morphology. By linking visible patterns to developmental events, live embryo staining supports studies of cell fate, organ formation, gene function, and developmental abnormalities while preserving the dynamics of the living system.

We describe a streamlined protocol for generating "fillet" preparations of Drosophila embryos of specific genotypes. This protocol allows efficient execution of a variety of genetic screens. It also allows excellent visualization of structures in the late embryo.

We introduce a novel hypoxic chamber system for use with aquatic organisms such as frog and zebrafish embryos. Our system is simple, robust, cost-effective and allows the induction and sustainment of hypoxia in vivo and for up to 48 h. We present 2 reproducible methods to monitor the effectiveness of hypoxia.
