Pcr Plate Centrifugation

PCR plate centrifugation is a brief, controlled spinning step used to bring reaction mixtures to the bottom of wells and prepare them for reliable polymerase chain reaction (PCR) amplification. During centrifugation, centrifugal force gathers droplets from well walls and lids, helps remove air bubbles, and promotes uniform contact among template DNA, primers, nucleotides, polymerase, and buffer; plates must be balanced to protect equipment and samples. In biology laboratories, this preparation improves volume consistency and reduces reaction variability before thermal cycling, supporting applications such as genotyping, cloning, pathogen detection, and gene-expression analysis. It is especially useful when liquid handling or sealing leaves droplets dispersed across the plate.

Working with Centrifuges

Source: Robert M. Rioux & Suprita Jharimune, Pennsylvania State University, University Park, PA Earth's gravitational force is capable of separating heterogeneous mixtures. However, many systems avert this type of separation owing to the length of time usually required in such cases. Centrifugation is one of the most powerful tools for the separation of heterogeneous mixtures1. It involves the application of centripetal force for sedimentation of the heavier phase, leading to separation of...

Centrifugation is a separation technique based on differences in density or size. It is commonly used to separate solids from aqueous interferents. During centrifugation, the sample is placed in centrifugation tubes and spun at high angular velocity, which allows centrifugal force to act differentially on the different densities or masses of the components. After spinning, the supernatant liquid is decanted. Depending on the specific application, either the pellet or the supernatant is retained...

An Introduction to the Centrifuge

The centrifuge is an instrument used in nearly every research lab across the globe. Centrifugation is the process by which a centrifuge is used to separate components of a complex mixture. By spinning laboratory samples at very high speeds, the components of a given mixture are subjected to centrifugal force, which causes more dense particles to migrate away from the axis of rotation and lighter ones to move toward it. These particles can sediment at the bottom of the tube into what’s...
