Positive Strand Rna

Positive-strand RNA is a type of single-stranded RNA whose sequence can function directly as messenger RNA, allowing genetic information to be translated into protein; it is important in molecular biology and the study of many RNA viruses. After entering a susceptible cell, the RNA is recognized by host ribosomes and translated into viral proteins, including an RNA-dependent RNA polymerase that copies the genome through a complementary negative-strand intermediate. This strategy supports rapid genome expression and replication, while its dependence on host translation machinery makes positive-strand RNA systems useful for investigating viral infection, antiviral drug targets, vaccine design, and RNA-based biotechnology.

Here, a protocol is presented for creating an infectious bacterial artificial chromosome containing a full-length cDNA of the positive-strand genomic RNA of Japanese encephalitis virus. This protocol can be used to construct a functional cDNA of other positive-strand RNA viruses, making it a powerful genomic tool for studying virus biology.

We describe an efficient method to separate single-stranded DNA, double-stranded DNA and RNA molecules from environmental viral communities. Nucleic acids are fractionated using hydroxyapatite chromatography with increasing concentrations of phosphate-containing buffers. This method permits the isolation of all viral nucleic acid types from environmental samples.

(2019)This video demonstrates the confocal imaging of viral double-stranded RNA (dsRNA) and cytoplasmic pattern recognition receptors (PRRs) in virus-infected epithelial cells. It highlights how antibody-based fluorescent labeling reveals the interaction between viral replication intermediates and host immune sensors.
