Post-thaw Survival

Post-thaw survival is the ability of cells, tissues, embryos, or other biological materials to remain viable and functional after cryopreservation and warming. During freezing, cryoprotectants, controlled cooling, and appropriate warming rates help limit ice-crystal formation, membrane damage, osmotic stress, and other injuries that can reduce viability. Assessing survival through membrane integrity, metabolic activity, recovery, or continued growth helps researchers optimize cryopreservation protocols and compare biological responses. In biology, post-thaw survival supports the storage and later use of cell lines, reproductive material, stem cells, tissues, and experimental samples for research, clinical applications, and biobanking.

Degassing Liquids with Freeze-Pump-Thaw Cycling

Source: Laboratory of Dr. Neil Branda — Simon Fraser University Degassing refers to the process by which dissolved gases are removed from a liquid. The presence of dissolved gases such as oxygen or carbon dioxide can impede chemical reactions that utilize sensitive reagents, interfere with spectroscopic measurements, or can induce unwanted bubble formation. A number of different techniques are available for degassing liquids; some of these include heating, ultrasonic agitation, chemical...

Since James Thomson et al developed a technique in 1998 to isolate and grow hES in culture, freezing cells for later use and thawing and expanding cells from a frozen stock have become important procedures performed in routine hES cell culture. Since hES cells are very sensitive to the stresses of freezing and thawing, special care must taken. Here we demonstrate the proper technique for rapidly thawing hES cells from liquid nitrogen stocks, plating them on mouse embryonic feeder cells, and...

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.
