Preclinical Animal Model

A preclinical animal model is a living nonhuman system used to study disease mechanisms, evaluate potential treatments, and assess safety before human trials. Researchers select an appropriate species and induce or reproduce relevant biological features, then measure responses to an intervention through clinical, physiological, behavioral, or molecular endpoints. These models can reveal therapeutic effects, toxicity, pharmacokinetics, and disease progression under controlled conditions. In biology, preclinical animal models support translational research by connecting laboratory findings with human health outcomes, while careful attention to model validity, reproducibility, and animal welfare helps determine how reliably results can inform clinical studies.

Here we provide a detailed procedure for large-scale production of research-grade AAV vectors using adherent HEK 293 cells grown in cell stacks and affinity chromatography purification. This protocol consistently yields >1 x 1013 vector genomes/mL, providing vector quantities appropriate for large animal studies.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.
