Reporter Gene Delivery

Reporter gene delivery is a molecular biology technique that introduces a reporter gene into cells to make gene expression or cellular activity detectable, often through fluorescence, luminescence, or enzymatic color change. Typically, researchers use plasmid transfection or viral transduction to place the reporter under a selected promoter; when that regulatory sequence is active, the cell produces a measurable reporter protein whose signal reflects promoter activity or pathway behavior. This approach supports studies of gene regulation, signaling, cell localization, and delivery efficiency, and can help evaluate experimental therapies, genetic constructs, and biological responses in cultured cells or model organisms.

Reporter genes are a type of protein-coding gene that are often tagged to a gene of interest. Once inside a target cell, reporter genes usually produce visually identifiable characteristics like fluorescence and luminescence when expressed along with the gene of interest. Thus, reporter genes “report” the presence or absence of genes of interest in an organism, determine the gene expression pattern, or track the physical location of a DNA segment or protein in the cell. Commonly used reporter...

Take a mixture of plasmids encoding hepatitis B surface antigens and lentiviral proteins, along with a plasmid containing a fluorescent reporter gene and a packaging signal.Add a transfection reagent, mix thoroughly, and incubate. The positively charged transfection reagent forms complexes with the negatively charged plasmid DNA.Introduce these complexes to a monolayer of human embryonic kidney cells and incubate. The complexes enter the cells via endocytosis.Replace the medium to remove residual...

Reporter Gene Repression Assay to Study Translational Regulation of a Target Gene

This video demonstrates an in vivo assay in bacteria to study the interaction of RNA-binding proteins (RBPs) with RNA. The bacterial cells are transformed with two plasmid constructs — a binding-site plasmid encoding an mRNA containing a fluorescent reporter gene downstream of an RBP-binding site — while an RBP plasmid expresses the RBP proteins under the control of an inducer. Upon inducer-mediated increase in RBP production, the RBPs bind to the mRNA, resulting in translational repression of...

Subpial Gene Delivery in the Mouse Spinal Cord to Evaluate Gene Expression

Source: Tadokoro, T., et al. Subpial Adeno-associated Virus 9 (AAV9) Vector Delivery in Adult Mice. J. Vis. Exp. (2017)This video demonstrates a subpial injection technique for gene delivery to the spinal cord in mice. The procedure involves exposing the dura mater, puncturing the pia mater, and injecting a recombinant adeno-associated virus (rAAV) carrying a fluorescent protein-expressing gene into the subpial space. Following injection and recovery, the virus facilitates gene expression in...

Implantation of cancer cells into the organ of origin can serve as a useful preclinical model to evaluate novel therapies. MB49 bladder carcinoma cells can be grown within the bladder following intravesical instillation. This protocol demonstrates catheterization of the mouse bladder for the purpose of tumor implantation and adenoviral delivery.
