Retinal Perfusion

Retinal perfusion is the delivery of oxygen and nutrients to retinal tissue through its blood supply, a process essential for vision and maintenance of neural cells. It depends on coordinated flow through the retinal and choroidal circulations, where vascular pressure, vessel diameter, and local metabolic demand regulate capillary exchange and removal of waste. Measuring retinal perfusion with techniques such as optical coherence tomography angiography, fluorescein angiography, or laser Doppler methods helps researchers assess vascular function without disrupting the eye. These measurements support the study of diabetic retinopathy, glaucoma, retinal vascular occlusions, and other conditions in which altered blood flow contributes to tissue damage or visual loss.

This article provides a description of how to dissect and record from the isolated retinal preparation in mouse. In particular, we describe how to record light responses from a fluorescently labeled ganglion cell population and subsequently identify and analyze its morphology.

Glutamate and Hypoxia as a Stress Model for the Isolated Perfused Vertebrate Retina

With this study, we introduce a standardized stress model for the isolated superfused bovine retina for future preclinical therapeutic testing. The effect of either hypoxia (pure N2) or glutamate stress (250 µM glutamate) on retinal function represented by a- and b-wave amplitudes was evaluated.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

Light-Evoked Postsynaptic Responses in Neurons of Mouse Retinal Slices

2025

This video demonstrates the recording of light-evoked post-synaptic responses in a dark-adapted mouse retinal tissue slice preparation. The retinal slice is placed in a recording chamber, and a recording pipette is used to establish electrical continuity with a retinal ganglion cell. Light pulses are applied to generate excitatory post-synaptic potentials in the ganglion cells, which are recorded by the pipette.
