Seed Culture Preparation

Seed culture preparation is the process of establishing a small, actively growing culture that provides a reliable inoculum for larger-scale biological cultivation. It typically involves transferring cells or microorganisms from a preserved or established stock into a suitable growth medium under aseptic conditions, then maintaining appropriate environmental conditions until the culture reaches a healthy growth phase. Careful preparation promotes consistent cell viability, reduces contamination risk, and supports reproducible scale-up. In biology, seed cultures are important for microbial cultivation, cell-based experiments, fermentation studies, and research requiring controlled growth or subsequent production in larger vessels.

2025

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.

Isolation of Tomato Seed-Associated Microbiota Through Controlled Germination and Comparative Culturing

2026

This protocol combines standardized seed surface sterilization with controlled germination to enable the reproducible isolation of tomato seed-associated microbiota during the seed-to-seedling transition. The approach enhances recovery of culturable microorganisms that are underrepresented in dry seeds and supports comparative culturomic studies during early seedling development.

Seed Structure and Early Development of the Sporophyte

2020

Seed structures are composed of a protective seed coat surrounding a plant embryo, and a food store for the developing embryo. The embryo contains the precursor tissues for leaves, stem, and roots. The endosperm and cotyledons—seed leaves—act as the food reserves for the growing embryo. The embryo contains a double set of chromosomes, one set from each parent. Fertilization of the haploid egg by the haploid sperm gives rise to the zygote, which develops into the embryo. The endosperm is a...

Chromosomes can be isolated from live cells such as lymphocytes or skin fibroblasts, and from organisms including humans or mice. These chromosome preparations can be further utilized for routine G-banding and molecular cytogenetic procedures such as fluorescence in situ hybridization (FISH), comparative genomic hybridization (CGH), and spectral karyotyping (SKY).
