Yeast Model

A yeast model is a living experimental system, most commonly Saccharomyces cerevisiae, used to investigate fundamental biological processes in a genetically tractable eukaryote. Researchers alter or monitor yeast genes and cells to examine mechanisms such as cell-cycle control, metabolism, protein folding, and stress responses, while conserved pathways can reveal principles relevant to more complex organisms. Rapid growth, well-characterized genetics, and controlled gene deletion or expression make yeast useful for studying gene function and disease-related processes. Findings from yeast models support research in molecular biology, biotechnology, and drug discovery, although differences from multicellular organisms require careful interpretation.

Yeast Maintenance

2023

Research performed in the yeast Saccharomyces cerevisiae has significantly improved our understanding of important cellular phenomona such as regulation of the cell cycle, aging, and cell death. The many benefits of working with S. cerevisiae include the facts that they are inexpensive to grow in the lab and that many ready-to-use strains are now commercially available. Nevertheless, proper maintenance of this organism is critical for successful experiments. This video will provide an overview...

Yeast Reproduction

2023

Saccharomyces cerevisiae is a species of yeast that is an extremely valuable model organism. Importantly, S. cerevisiae is a unicellular eukaryote that undergoes many of the same biological processes as humans. This video provides an introduction to the yeast cell cycle, and explains how S. cerevisiae reproduces both asexually and sexually Yeast reproduce asexually through a process known as budding. In contrast, yeast sometimes participate in sexual reproduction, which is important because...

2010

Genetic studies in yeast can be employed to investigate the molecular and cellular functions of human genes in cellular DNA metabolism. Methods are described for the genetic characterization of the human WRN gene product defective in the premature aging disorder Werner syndrome in functionally conserved pathways using yeast as a tractable model system.

Yeast Transformation and Cloning

2023

S. cerevisiae are unicellular eukaryotes that are a commonly-used model organism in biological research. In the course of their work, yeast researchers rely upon the fundamental technique of transformation (the uptake of foreign DNA by the cell) to control gene expression, induce genetic deletions, express recombinant proteins, and label subcellular structures. This video provides an overview of how and why yeast transformation is carried out in the lab. The important features of yeast...

Yeast Two-Hybrid Assay to Determine Protein Self-Association in Yeast Cells

2025

This video demonstrates the yeast two-hybrid assay to detect specific protein self-interactions. In this assay, the protein of interest is tagged to two domains of a transcription factor that encodes the β-galactosidase enzyme. When the two proteins interact, this causes transcription and translation to form the enzyme, which is then detected using a biochemical assay.
