Arrayed Lentiviral Libraries

Arrayed lentiviral libraries are collections of lentiviral vectors, each carrying a defined genetic element and stored in a known well, enabling systematic gene studies without the ambiguity of pooled screens. Each vector enters susceptible cells, and lentiviral machinery delivers its cargo for stable integration into the host-cell genome, producing a reproducible perturbation such as gene suppression or expression. In cancer research, these libraries support parallel testing of genes, regulatory sequences, or therapeutic targets across tumor cell models. The resulting phenotypes, including changes in proliferation, survival, or drug response, help identify cancer dependencies, validate targets, and clarify mechanisms of treatment resistance.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.

Here we use a human LentiPlex pooled library and traditional sequencing methods to identify gene targets promoting cell survival. We demonstrate how to set up and deconvolute a LentiPlex screen and validate the results.
