Eosin Y Counterstain

Eosin Y counterstain is an acidic histological dye used to add contrast to tissue sections after a primary stain, making cellular and tissue architecture easier to interpret. In hematoxylin and eosin (H&E) staining, negatively charged eosin binds positively charged proteins, particularly in cytoplasm, extracellular matrix, and erythrocytes, producing shades from pink to red while hematoxylin highlights nuclei blue-purple. In cancer research, this contrast supports assessment of tumor morphology, cellular atypia, stromal organization, necrosis, and inflammatory features in biopsy and model tissues. Eosin Y counterstaining remains a foundational method for routine pathology, disease characterization, and correlation with molecular or immunohistochemical findings.

Hematoxylin and Eosin Staining to Assess Colon Damage in DSS-Induced Colitis

2025

This video demonstrates the hematoxylin and eosin (H&E) staining method to evaluate colon damage in a murine model of DSS-induced colitis — an inflammatory bowel disease. Upon isolating the colon from a DSS-treated mouse and obtaining sections of the tissue, H&E staining of the sections helps identify the inflammation-mediated tissue damage.

H&E Staining of Paraffin-Embedded Tissue Sections: A Differential Staining Technique to Visualize Liver Tissue Sections Using Combination of Hematoxylin and Eosin Dyes

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
