Mesenteric Lymphatics

Mesenteric lymphatics are vessels and lymphoid structures within the mesentery that drain fluid, lipids, immune cells, and antigens from the intestines to the lymphatic circulation. Blind-ended lymphatic capillaries collect interstitial fluid, while larger collecting vessels use valves and rhythmic smooth-muscle contractions to propel lymph toward regional lymph nodes and the thoracic duct. In cancer research, this network is important because tumor cells from gastrointestinal and abdominal cancers can enter lymphatic vessels, travel to mesenteric or regional nodes, and establish metastases. Studying mesenteric lymph flow and lymph node involvement supports research into tumor spread, staging, immune regulation, and targeted therapies.

The Mesenteric Lymph Duct Cannulated Rat Model: Application to the Assessment of Intestinal Lymphatic Drug Transport

Here we describe a technique to cannulate the mesenteric lymph duct in rats which enables quantification of lipid and drug transport via the lymphatic system following intestinal delivery. The technique can be adapted to assess mesenteric lymph concentrations and/or transport of fluid, immune cells, peptides, proteins and lipophilic molecules.

Angiogenesis involves multi-cell, multi-system interactions that need to be investigated in a physiologically relevant environment. The objective of this study is to demonstrate the ability of the rat mesentery culture model to make time-lapse comparisons of intact microvascular networks during angiogenesis.

This experimental protocol describes a hepatic lymph duct-cannulated rat model that enables direct and quantitative assessment of hepatic lymphatic and vascular drainage. The model allows simultaneous sampling of hepatic lymph and systemic blood to investigate hepatic transport, metabolism, and immune signaling, supporting studies of drug pharmacokinetics and liver-specific biology.
