Imine Derivative Formation

Imine derivative formation is a condensation reaction that converts aldehydes or ketones and primary amines into compounds containing a carbon–nitrogen double bond, known as imines or Schiff bases. The process proceeds through nucleophilic addition of the amine to the carbonyl group, followed by proton transfers and elimination of water; mild acid catalysis and removal of water can promote product formation. In chemistry, this reaction provides a practical route for preparing intermediates, ligands, dyes, and biologically relevant molecules. Imine formation also supports carbonyl compound identification and demonstrates how reversible equilibrium and functional-group reactivity guide synthetic design.

Primary amines react with carbonyl compounds—aldehydes and ketones—to generate imines. Imines consist of a C=N double bond and are named Schiff bases after its discoverer—the German chemist Hugo Schiff. On the other hand, secondary amines react with carbonyl compounds to give enamines. In enamines, the presence of a C=C double bond adjacent to the nitrogen atom leads to the delocalization of the lone pair. Both imine formation and enamine formation are reversible and acid-catalyzed.

The aim of this method is to obtain fibroblast-derived 3D matrices as a natural scaffold for subsequent cellular assays. Fibroblasts are seeded in a pre-treated culture plate and stimulated with ascorbic acid for matrix generation. Matrices are decellularized and blocked to culture relevant cells (e.g., endothelial cells).
