Endocervical Brush

An endocervical brush is a slender, soft-bristled sampling device used to collect epithelial cells and secretions from the endocervical canal, supporting clinical evaluation of cervical health. During specimen collection, the brush is guided through the cervical opening and rotated so its bristles contact the canal lining; the collected material is then transferred to a slide or liquid-based preservative for laboratory analysis. Endocervical brushes are commonly used with cervical cytology and human papillomavirus testing to help identify abnormal cellular changes and infection-related risk. Proper sampling improves specimen adequacy, strengthening screening and diagnostic workflows in women’s health.

Isolation and Flow Cytometric Analysis of Human Endocervical Gamma Delta T Cells

We describe here an isolation method to obtain human endocervical intraepithelial lymphocytes for the analysis of intraepithelial gamma delta T cells. This protocol can be extended for the purification of endocervical gamma delta T cells by magnetic beads or by cell sorting.

Preparation and Friction Force Microscopy Measurements of Immiscible, Opposing Polymer Brushes

The methodology to perform friction force microscopy experiments for contacting brushes is presented: Two polymer brushes that are grafted from (a) substrates and (b) colloidal probes are slid to show that, by using two contacting immiscible brush systems, friction in sliding contacts is reduced compared to miscible brush systems.

Poly(ethylene glycol) (PEG) brush-arm star polymers (BASPs) with narrow mass distributions and tunable nanoscopic sizes are synthesized in via ring opening metathesis polymerization (ROMP) of a PEG-norbornene macromonomer followed by transfer of portions of the resulting living brush initiator to vials containing varied amounts of a rigid, photo-cleavable bis-norbornene crosslinker.

The use of cytobrush sampling to collect lymphocytes and monocytes from the endocervix is a minimally invasive technique that provides samples for analysis of female genital tract immunity. In this protocol, we describe the collection of cytobrush samples and immune cell isolation for flow cytometry assays.

Brush cells are rare cholinergic chemosensory epithelial cells found in the naïve mouse trachea. Due to their limited numbers, ex vivo evaluation of their functional role in airway immunity and remodeling is challenging. We describe a method for isolation of tracheal brush cells by flow cytometry.
