Leak Test

A leak test is a method for detecting unintended openings or loss of containment in a component, package, or fluid pathway, helping verify integrity and safety in clinical settings. The test typically applies a controlled pressure or vacuum and monitors pressure change over time, while some methods detect the escape of a tracer gas or liquid through a defect. Leak testing supports quality control for sterile medical packaging, tubing, catheters, syringes, and other devices that must contain fluids or gases. Reliable results can identify manufacturing defects, reduce contamination risk, and confirm performance before clinical use.

The endoplasmic reticulum plays a key role in protein biogenesis and in calcium homeostasis. We have established an experimental system that allows us to address the role of Ca2+ leak channels and to characterize their putative regulatory mechanisms. This system involves siRNA mediated gene silencing and live cell Ca2+ imaging.

Here, we share methods for measuring mitochondrial oxygen consumption, a defining concept of nutritional energetics, and proton leak, the primary cause of inefficiency in mitochondrial generation of ATP. These results can account for 30% of the energy lost in nutrient utilization to help evaluate mitochondrial function.

Viability Assays for Cells in Culture

Therapeutic compounds are often first examined in vitro with viability assays. Blind cell counts by a human observer can be highly sensitive to small changes in cell number but do not assess function. Computerized viability assays, as described here, can assess both structure and function in an objective manner.
