Tablet Formulation

Tablet formulation is the design and manufacture of solid oral dosage forms that deliver an active pharmaceutical ingredient accurately, consistently, and in a stable format. It combines the drug with excipients such as binders, fillers, disintegrants, and lubricants, then uses processes including blending, granulation, compression, and coating to control tablet strength, dissolution, and handling. In clinical development, formulation choices influence bioavailability, dosing reliability, patient acceptability, and shelf life. Well-designed tablets support reproducible drug release and scalable production, while specialized formulations can modify release rates or protect sensitive ingredients from the gastrointestinal environment.

Formulation and Manufacturing Process: Physical Attributes of Generic Tablets and Capsules

2025

Bioequivalence in generic drugs, such as tablets and capsules, refers to their pharmaceutical equivalence to the brand-name counterparts. However, for therapeutic equivalence, manufacturers must also consider physical attributes like size, shape, and weight (FDA Guidance for Industry, December 2003). Discrepancies in these aspects could impact patient compliance and cause medication errors. For instance, swallowing difficulties, often experienced with larger tablets or capsules, can lead to...
