Cerebral Injury

Cerebral injury is damage to the brain caused by trauma, disrupted blood flow, infection, toxic exposure, or other insults, and it can alter neural structure and function across the lifespan. After the initial damage, impaired energy metabolism and blood-brain barrier disruption can trigger inflammation, excitotoxic signaling, oxidative stress, and cell death, while immature neural circuits may respond differently during development. In developmental biology, studying cerebral injury helps researchers determine how the timing and severity of damage affect neurogenesis, neuronal migration, circuit formation, and functional recovery. Experimental models and imaging approaches support evaluation of protective therapies and strategies that promote repair, compensation, and healthy brain development.

Non-invasive Optical Measurement of Cerebral Metabolism and Hemodynamics in Infants

We combined frequency-domain near-infrared spectroscopy measures of cerebral hemoglobin oxygenation with diffuse correlation spectroscopy measures of cerebral blood flow index to estimate an index of oxygen metabolism. We tested the utility of this measure as a bedside screening tool to evaluate the health and development of the newborn brain.

Focal Cerebral Ischemia Model by Endovascular Suture Occlusion of the Middle Cerebral Artery in the Rat

Surgical induction of ischemic brain damage in the rat is a widely used model for stroke research. Here we demonstrate the induction of focal cerebral ischemia by occlusion of the middle cerebral artery. Visualization of the resulting infarct by histological staining and magnetic resonance imaging is also shown.

Controlled Cervical Laceration Injury in Mice

A novel technique to create a reproducible in vivo model of cervical spinal cord laceration injury in the mouse is described. This technique is based on spine stabilization by fixation of the cervical facets and laceration of the spinal cord using an oscillating blade with an accuracy of ±0.01 mm.

A Battery of Motor Tests in a Neonatal Mouse Model of Cerebral Palsy

Presented is a concise battery of mouse neonatal motor tests. Using these tests, neonatal motor deficits can be demonstrated in a variety of neonatal motor disorders. By having a standardized set of tests, results from different studies can be compared, allowing for better and accurate reporting between groups.

Isolation and Cannulation of Cerebral Parenchymal Arterioles

This manuscript describes a simple and reproducible protocol for isolation of intracerebral arterioles (a group of blood vessels encompassing parenchymal arterioles, penetrating arterioles and pre-capillary arterioles) from mice, to be used in pressure myography, immunofluorescence, biochemistry, and molecular studies.
