Endothelial To Hematopoietic

Endothelial-to-hematopoietic transition is a developmental process in which specialized hemogenic endothelial cells generate blood-forming cells, establishing the embryonic hematopoietic system. During this transition, endothelial cells in the arterial vasculature change shape and identity, activate regulators such as RUNX1 and GATA2, and bud from the vessel wall into the lumen as hematopoietic stem and progenitor cells. Studying this process clarifies how blood development is initiated and coordinated with vascular formation. It also informs efforts to produce blood stem cells from pluripotent stem cells, model congenital blood disorders, and improve regenerative and transplantation strategies.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

Here, we present human pluripotent stem cell (hPSC) culture protocols, used to differentiate hPSCs into CD34+ hematopoietic progenitors. This method uses stage-specific manipulation of canonical WNT signaling to specify cells exclusively to either the definitive or primitive hematopoietic program.

We have identified the placenta as a major hematopoietic organ during development. We found that hematopoietic stem cells (HSCs) are both generated and expanded in the placenta in unique microenvironmental niches. Here, we describe experimental techniques required for isolation and visualization of HSCs in the mouse placenta.
