Ex Vivo Droplet Culture

Ex vivo droplet culture is a method for maintaining isolated cells, tissues, or developing organ structures outside the organism in small, defined droplets of culture medium. Each droplet provides a controlled three-dimensional microenvironment that supports nutrient and gas exchange while preserving cell-to-cell interactions, tissue organization, and developmental signaling. In developmental biology, researchers use this approach to observe morphogenesis, test how genes or signaling pathways regulate tissue formation, and assess responses to experimental compounds under defined conditions. Because droplets require little sample volume and permit direct imaging, they enable efficient analysis of developmental processes while reducing dependence on whole-animal experiments.

The ex vivo upright droplet culture is an alternative to current in vitro and in vivo experimental techniques. This protocol is easy to perform and requires smaller amounts of reagent, while permitting the ability to manipulate and study fetal vascularization, morphogenesis, and organogenesis.

A System for ex vivo Culturing of Embryonic Pancreas

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

This video demonstrates New culture, a method by which chick embryos are cultured outside the egg for up to 24 hr. This method enables one to study early development (primitive streak to 14 som.), a period corresponding to E7-9 in mouse. Applications of this technique include electroporation, in situ hybridization and immunohistochemistry.
