Lymphoid Progenitor Expansion

Lymphoid progenitor expansion is the controlled increase of lymphoid precursor cells that give rise to B cells, T cells, natural killer cells, and other immune populations, enabling researchers to study how the immune system develops. In culture, defined combinations of growth and survival signals, including stem cell factor, FLT3 ligand, and interleukin-7, promote progenitor maintenance and proliferation, while Notch signaling can bias cells toward T-lineage development. This approach helps reveal how cellular signals regulate lymphoid commitment and differentiation. It also supports developmental biology research, immune-cell production, disease modeling, and evaluation of strategies for regenerative medicine and hematopoietic therapies.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

2025

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.

Endothelial colony forming progenitor cells (ECFCs) are a promising tool to study vascular homeostasis and repair.1,2 This paper introduces a novel animal-serum free method for isolation and expansion of ECFC from heparinised adult human peripheral blood with pooled human platelet lysate (pHPL) diminishing the risk of anti-bovine immunisation.

Here we report a method for isolation of Adipocyte Progenitor Cell (APC) populations from Perivascular Adipose Tissue (PVAT) using Magnetic-activated Cell Sorting (MCS). This method allows for an increased isolation of APC per gram of adipose tissue when compared to Fluorescence-Activated Cell Sorting (FACS).
