Mesoderm Lineage Induction

Mesoderm lineage induction is the developmental process that directs pluripotent or early embryonic cells to form mesoderm, the germ layer that produces muscle, bone, blood, connective tissue, and much of the urogenital system. During gastrulation, coordinated signaling by pathways such as Nodal/Activin, Wnt, and bone morphogenetic proteins establishes mesodermal identity and guides cells toward specific positional and tissue fates. In developmental biology, researchers study these signals using embryos, stem cell cultures, and organoid models to define gene regulatory networks and the timing of lineage decisions. Controlled mesoderm induction also supports disease modeling, tissue engineering, and the generation of differentiated cells for regenerative research.

Here, we present a protocol for a mosaic labeling technique that permits the visualization of neurons derived from a common progenitor cell in two distinct colors. This facilitates neural lineage analysis with the capability of birth-dating individual neurons and studying gene function in the same neurons of different individuals.

Somitogenesis is a rhythmic developmental process that spatially patterns the body axis of vertebrate embryos.
