Neural Stem Cell Expansion

Neural stem cell expansion is the controlled increase of neural stem cell numbers while preserving their capacity for self-renewal and differentiation into neurons, astrocytes, and oligodendrocytes. In culture, mitogens such as epidermal growth factor and fibroblast growth factor 2 stimulate proliferation, while defined media and controlled passage conditions help maintain an undifferentiated state; withdrawing these signals can initiate differentiation. This method enables researchers to generate sufficient cells for studying neural development, lineage specification, and neurogenesis. Expanded neural stem cells also support disease modeling, drug testing, and investigations of cell-based strategies for nervous system repair.

Controlling the expansion of somatic stem cells is a major factor hampering their study and use in therapy. Here we describe a system to temporally control neural stem cells expansion during development and adulthood, which can be used to increase the number of neurons generated in the mouse brain.
