Primary Islet Handling

Primary islet handling refers to the controlled collection, transfer, culture, and experimental manipulation of freshly isolated pancreatic islets while preserving their structure and viability. Because islets are delicate three-dimensional clusters, gentle pipetting, appropriate culture conditions, and limited mechanical stress help maintain cell-cell interactions and endocrine function. In developmental biology, careful handling supports studies of islet formation, endocrine-cell differentiation, and maturation by allowing researchers to examine intact tissue or prepare it for imaging, molecular analysis, and functional assays. Consistent handling also improves reproducibility when comparing developmental stages or testing factors that influence pancreatic islet development.

This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.

Here, we present a protocol to guide human primary prostate organoid handling then suggest endpoints to assess phenotype. Seeding, culture maintenance, recovery from matrix gel, morphologic quantification, embedding and sectioning, FFPE sectioning, whole-mount staining, and application of commercial assays are described.
